The following 38 teams from 11 associations (the 10 CONMEBOL members plus Mexico which were invited to compete) qualified for the tournament:
- Title holders
- Argentina and Brazil: 5 berths each
- All associations other than Argentina and Brazil: 3 berths each
Among them, 12 teams (the team with the lowest berth from each of the 11 associations, plus the team with the second lowest berth from the association of the title holders) are entered in the first stage, and the remaining 26 teams are entered in the second stage.
- Teams which have qualified, but not certain of specific berth
- Paraguay (PAR): Cerro Porteño (2015 Apertura champion) may earn the Paraguay 1 or Paraguay 2 berth.
- Notes
- Title holders (TH): The 2015 Copa Libertadores champion will qualify for the second stage, meaning the team with the second lowest berth from the association of the title holders will qualify for the first stage instead.
- Copa Sudamericana winners (SUD): The 2015 Copa Sudamericana champion are guaranteed to qualify for the tournament. If they have not otherwise qualified according to the internal distribution method of their association, they will take this berth instead of the team designated here.
- Mexico (MEX): Ordinarily, the winner of the 2015 Supercopa MX, initially to be played between Santos Laguna (2014 Apertura Copa MX champion) and Puebla (2015 Clausura Copa MXchampion), earns the Mexico 3 berth. However, Santos Laguna is ineligible for South American competition due to their participation in the 2015–16 CONCACAF Champions League, and also will not play in the 2015 Supercopa MX (as they won the 2015 Clausura and will play in the 2015 Campeón de Campeones on the same night), where they are replaced by Morelia (2013 Apertura Copa MX champion). So Puebla will qualify automatically for the 2016 Copa Libertadores regardless of whether they win or lose in the 2015 Supercopa MX.
